The question or problem should be grounded on information from previous units. It should require the learner to apply basic information from the course in a higher order of thinking (i.e. the answer should not be attainable purely from the text or lectures) For example (please do not use these examples): In an experiment, a valine is mutated to a glutamic acid in a transmembrane domain of a protein. What is the likely consequence of this single amino acid change? A new potential analog of cortisol is being synthesized that is not membrane permeable. Response: Based on information in unit 1, glutamic acid is a polar amino acid and the introduction of a charge into a hydrophobic environment (the membrane) would disrupt the membrane structure. This may also reduce the stability of the protein in the membrane environment.
