Virtual Urchin Lab: Analyzing Gene Function
Objective: Virtually, use a program provided by The University of Washington to
examine skeleton formation in sea urchin larva. Use morpholinos to interfere with gene
expression (a Lose It experiment!) and function to determine how the sea urchin
skeleton is impacted.
Procedure: Visit the following website:

Once you reach this page, under Interactive Tutorials ? 2. Development and
Embryology, click on “analyzing gene function.” If needed, This will take you to
the main page where the experiment is broken down into 4 parts. You need to
complete each part of this virtual exercise, as the questions in the post-lab
assignment are taken from all four parts of the lab. Read all components of the
lab, follow the directions, and answer any questions that the site provides you
with. The questions for the post-lab assignment are found below:
Total of 50 points
Answer all questions on a separate sheet of paper. All questions must be
typed up.
For any questions requiring an explanation, please use complete sentences,
proper grammar and spelling, and organize your answers in a concise and logical
manner. Points will be deducted for such errors. You MUST answer in your
own words – simply copying from the website for any question will result in
zero points awarded. If your assignment is entirely plagiarized, you will
receive a zero on the whole exercise.
1.. Why is it imperative to use a needle puller to prepare the needles for the sea
urchin egg injections? (2 points)
2. Just by looking under the microscope you should be able to tell if an egg has
been fertilized or not. What will you specifically look for in order to make that
determination? (1 point)
3. Suppose that you row or line up your eggs and then you use the
micromanipulator to move the needle into the egg. You successfully inject one of
your morpholinos into the egg and then immediately move onto the next egg.
Explain what did you do wrong (2 points).
4.. In the context of the purpose of this experiment why are we measuring arm
length in the sea urchins that developed from the eggs we injected? (3 points)
5.. How did you distinguish between the control and non-injected groups? What
kind of light did you use? Explain your answers (5 points).
6.. In your own words, what are the two main questions that we are asking in
the statistical analysis section? (2 points)
7.Click on the full dataset to display the statistics and explain what the generated
results mean. Are there any statistically significant results? Relate your answer
back to the two main questions that you posed above (5 points).
8. After completing this exercise, you sit down with your lab partner and discuss
the results. She explains that we now know that Alx1 is the molecule responsible
for forming the calcium carbonate skeleton in sea urchins. Is she correct? Explain
your answer (3 points).
